چکیده

The enzyme L-Amino Acid Oxidase (LAAO) is widely distributed in snake venoms, contributes a reasonable toxicity to the venoms. However LAAO from Indian cobra (Naja naja ) snake venom has not yet isolated previously. In the present study LAAO from Indian cobra (Naja naja ) snake venom was purified by sequential steps of cation exchange chromatography (CM-Sephadex C-25; CM-52 cellulose), followed by sephadex G-100 gel filtration chromatography. The purified enzyme was named as Indian Cobra Naja naja LAmino Acid Oxidase (ICN-LAAO). This ICN-LAAO is a monomer and its molecular mass has been found to be 61.91 kDa determined by MALDI-TOF and 12% SDS-PAGE. The enzyme has an isoelcetric point of 8.12 and a pH optimum of 8.5. It shows remarkable thermal stability and partial inactivation by freezing. The enzyme may contribute in the development of severe hematological disorder due to cobra venom envenomation.

Homology Modeling and Prediction of Catalytic Amino Acid in the Neurotoxin from Indian Cobra (Naja naja)

The neurotoxin secreted by Indian cobra (Naja naja) binds to acetylcholine receptor of nerve cells, which leads to a lump in the nerve impulse, ceases breathing, thereby causing the death of a person due to suffocation. These neurotoxins are small peptide with approximately 7 kDa. Homology modeling was performed to generate the 3D structure of neurotoxins (A, B, C and D) of N. naja, using the k...

The basic proteins of cobra venom. I. Isolation and characterization of cobramines A and B.

Cobra venoms contain heat stable, basic, proteins that inhibit iodide accumulation by thyroid slices. Activity is not related to the content of phospholipase A or various other enzymatic activities. These proteins are present only in Elapid venoms and not in snakes of the Crotalidae or Viperidae families. Two such basic proteins, named cobramines A and B, have been purified from the crude venom...

Snake venom toxins. The amino acid sequences of toxins b and d from Naja melanoleuca venom.

Two toxins have been isolated from the venom of the Forest cobra (Naja melanoleuca) and their amino acid sequences elucidated. Toxin b, a 71 amino acid residue molecule, is devoid of methionine and resembles toxin (x from Naja nivea venom both immunochemically and in amino acid sequence. Toxin d consists of 61 amino acid residues with alanine, phenylalanine, and leucine totally absent. Toxin d ...

Identification of serotonin-sensitive aryl acylamidase activity with cobra venom acetylcholinesterase.

Acetylcholinesterase purified from cobra (Naja naja) venom exhibits a serotonin-sensitive aryl acylamidase activity. Both acetylcholinesterase and aryl acylamidase activities co-eluted in column chromatographic procedures (Sephadex G-75 and Zinc-Sepharose), co-migrated on polyacrylamide gel electrophoresis, co-immunoprecipitated by anti-snake venom antibody and showed the same heat denaturation...

Hematology and serum biochemistry of Indian spectacled cobra (Naja naja) and Indian rat snake (Ptyas mucosa)

AIM To study the hematology and serum biochemistry parameters of Indian spectacled cobra (Naja naja) and Indian rat snake (Ptyas mucosa) and to evaluate the differences in the same between captive and wild populations. MATERIALS AND METHODS Animals were categorized into four groups, viz., wild Indian spectacled cobra (n=10), wild Indian rat snakes (n=10), captive Indian spectacled cobra (n=10...
